The IDH3 alpha (IDH3α) antibody is a crucial tool in studying the function and expression of isocitrate dehydrogenase 3 alpha, a subunit of the IDH3 enzyme complex. IDH3 is a key metabolic enzyme in the tricarboxylic acid (TCA) cycle, catalyzing the oxidative decarboxylation of isocitrate to α-ketoglutarate while generating NADH. Unlike its isoforms IDH1 and IDH2. which are frequently mutated in cancers (e.g., gliomas, AML), IDH3α mutations are rare but have been implicated in certain metabolic disorders and cancer subtypes. The IDH3α subunit, encoded by the IDH3A gene, forms the catalytic core of the heterotrimeric IDH3 complex (α/β/γ), essential for mitochondrial energy production.
Antibodies targeting IDH3α are widely used in research to investigate its expression patterns, subcellular localization (mitochondrial), and dysregulation in diseases. These antibodies are validated for applications such as Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF). Given the structural similarities among IDH family members, high specificity is critical for distinguishing IDH3α from IDH1/IDH2. Some antibodies are designed to detect post-translational modifications or mutations linked to functional alterations. In cancer research, IDH3α antibodies help explore metabolic reprogramming ("Warburg effect") and potential therapeutic targets. Additionally, they aid in studying rare IDH3A-associated genetic disorders, such as retinitis pigmentosa and enzyme deficiency syndromes. Proper validation using knockout controls or recombinant proteins is essential to ensure antibody reliability, as off-target binding can complicate interpretation.
