背景信息

The CALY (N-term) antibody is a polyclonal or monoclonal antibody specifically designed to target the N-terminal region of the Calcyon protein (CALY), a neuron-specific transmembrane protein implicated in synaptic signaling and intracellular trafficking. Calcyon interacts with dopamine D1 receptors and other synaptic proteins, modulating dopamine-mediated neurotransmission and clathrin-associated endocytosis. Its expression is enriched in the prefrontal cortex, hippocampus, and striatum, regions critical for cognitive and emotional processing. Dysregulation of Calcyon has been linked to neuropsychiatric disorders, including schizophrenia and attention-deficit/hyperactivity disorder (ADHD), making it a research focus in neuroscience and psychiatry.

The N-terminal antibody is commonly generated in rabbits or mice using synthetic peptides or recombinant proteins corresponding to the first 50-100 amino acids of Calcyon. It is validated for applications such as Western blotting, immunohistochemistry, and immunofluorescence to detect endogenous Calcyon expression in brain tissues or cultured neurons. Researchers utilize this antibody to study Calcyon's subcellular localization, protein-protein interactions, and regulatory roles in dopamine signaling pathways. Specificity is often confirmed via knockout controls or peptide blocking assays. Its development has advanced understanding of Calcyon's dual role in clathrin-coated vesicle formation and dopamine receptor trafficking, offering insights into synaptic plasticity mechanisms and potential therapeutic targets for dopamine-related disorders.
