背景信息

CD34 is a transmembrane glycoprotein primarily expressed on hematopoietic stem/progenitor cells, endothelial cells, and certain mesenchymal stromal cells. First identified in the 1980s, the CD34 antigen became a critical marker for isolating human hematopoietic stem cells (HSCs) using monoclonal antibodies. The CD34 gene encodes a highly glycosylated single-pass surface protein (110–120 kDa) involved in cell adhesion, differentiation, and vascular remodeling.

CD34 antibodies were initially developed through hybridoma technology, with clones like QBEND10 and My10 becoming widely used. These antibodies target specific epitopes on the extracellular domain, enabling applications in flow cytometry, immunohistochemistry, and immunomagnetic cell sorting. Clinically, CD34+ cell enumeration guides stem cell transplantation and leukemia diagnosis, as aberrant CD34 expression correlates with certain malignancies.

Despite its historical importance, CD34’s biological role remains incompletely understood. It may mediate cell-cell interactions via ligands like L-selectin and regulate stem cell homing. Controversies persist about its exclusivity as a stem cell marker, as some functional HSCs are CD34-.
