UHRF2 (Ubiquitin-like with PHD and RING Finger domains 2) is a multifunctional protein belonging to the UHRF family, which also includes UHRF1. It plays a role in epigenetic regulation, particularly in maintaining DNA methylation patterns and histone modification states. Structurally, UHRF2 contains ubiquitin-like (UBL), tandem Tudor, plant homeodomain (PHD), SET- and RING-associated (SRA), and RING finger domains, enabling interactions with histones (e.g., H3K9me3), DNA, and chromatin-modifying enzymes. Unlike UHRF1. which is well-studied in DNA replication and methylation, UHRF2’s functions are less defined but involve cell cycle regulation, DNA damage repair, and ubiquitination processes.
Research links UHRF2 to cancer, neurodevelopment, and metabolic diseases. It exhibits dual roles in tumors—acting as an oncogene or tumor suppressor depending on context—with altered expression noted in hepatocellular carcinoma (upregulated) and colorectal cancer (downregulated). UHRF2 antibodies are critical tools for studying its expression, localization, and interactions in vitro and in vivo. They are widely used in techniques like Western blotting, immunofluorescence, and chromatin immunoprecipitation. Specificity validation (e.g., knockout controls) is essential due to structural similarities within the UHRF family. Ongoing studies aim to clarify UHRF2’s mechanistic roles and therapeutic potential in disease.
