The PAX5 antibody is a crucial tool in immunology and pathology for identifying the PAX5 protein, a transcription factor encoded by the PAX5 gene. PAX5. part of the paired box (PAX) family, is essential for B-cell development, regulating genes involved in lineage commitment, differentiation, and maturation. It maintains B-cell identity by activating B-cell-specific genes (e.g., CD19. BLK) and repressing genes associated with alternative lineages. PAX5 expression is predominantly observed in B-lymphoid cells, from pro-B to mature B-cell stages, but is absent in plasma cells and non-B-cell lineages.
In diagnostic pathology, PAX5 antibodies are widely used in immunohistochemistry (IHC) to confirm B-cell lineage in malignancies, such as B-cell acute lymphoblastic leukemia (B-ALL) and B-cell lymphomas (e.g., diffuse large B-cell lymphoma). Its nuclear staining pattern helps distinguish B-cell neoplasms from T-cell or myeloid malignancies. However, PAX5 expression loss or mutation is linked to aggressive B-ALL subtypes and lymphomagenesis.
PAX5 antibodies also aid in research exploring B-cell development, differentiation defects, and PAX5’s role in tumor suppression. Aberrant PAX5 expression is implicated in autoimmune diseases and solid tumors. While highly specific for B-cells, cross-reactivity with other PAX family proteins in non-B tissues requires careful interpretation. Overall, PAX5 antibodies remain indispensable for both clinical diagnostics and molecular studies of B-cell biology and pathology.
