ULK1 (Unc-51 like autophagy activating kinase 1) is a serine/threonine-protein kinase central to autophagy initiation in mammalian cells. Evolutionarily conserved, ULK1 homologs like yeast Atg1 regulate autophagy—a lysosome-dependent degradation process critical for cellular homeostasis. ULK1 operates within a multiprotein complex, interacting with ATG13. FIP200. and ATG101. which collectively respond to cellular stress signals such as nutrient deprivation.

Its activity is tightly regulated by post-translational modifications. Under nutrient-rich conditions, mechanistic target of rapamycin complex 1 (mTORC1) phosphorylates ULK1. suppressing its kinase activity.
