Ubiquitin-like with PHD and RING Finger Domains 1 (UHRF1), also known as NP95 or ICBP90. is a multidomain epigenetic regulator crucial for maintaining DNA methylation patterns during cell division. By bridging DNA methylation and histone modifications, it ensures the faithful inheritance of epigenetic information. Structurally, UHRF1 contains a ubiquitin-like domain (UBL), tandem Tudor domain (TTD), plant homeodomain (PHD), SET- and RING-associated (SRA) domain, and a RING-type E3 ubiquitin ligase domain. The SRA domain recognizes hemi-methylated DNA through 5-methylcytosine, while its TTD and PHD domains interact with unmodified histone H3 tails, preferentially binding to H3K9me0 and H3K4me0. This dual recognition mechanism couples DNA replication with the restoration of repressive histone marks. UHRF1’s RING domain recruits DNMT1 to replication foci, facilitating methylation of newly synthesized DNA.
