CD46. also known as membrane cofactor protein (MCP), is a transmembrane regulatory protein widely expressed on nucleated human cells. It belongs to the complement regulatory protein family and plays a critical role in preventing unintended complement-mediated damage to host cells by binding to complement proteins C3b and C4b, facilitating their inactivation. Beyond its immune regulatory function, CD46 serves as a receptor for several pathogens, including measles virus, human herpesvirus 6. and certain adenoviruses, enabling their cellular entry.
CD46 antibodies are tools or therapeutics targeting this protein. In research, they are used to study CD46's roles in immune modulation, viral pathogenesis, and cell signaling. Clinically, CD46 has gained attention due to its overexpression in various cancers, making it a potential therapeutic target. Antibodies against CD46 are explored in cancer immunotherapy, either to block pro-survival signals in tumors or to deliver cytotoxic agents selectively. Additionally, CD46 antibodies have implications in autoimmune diseases and reproductive medicine, given its involvement in sperm-egg interaction and pregnancy-related immune tolerance.
The development of CD46-specific monoclonal antibodies (e.g., M177) has advanced both diagnostic and therapeutic applications. Challenges include managing CD46's ubiquitous expression to minimize off-target effects. Ongoing studies aim to refine antibody specificity and explore bispecific designs to enhance therapeutic efficacy in oncology and infectious diseases.
