背景信息

The tumor protein p73 (TP73), a member of the p53 family, plays a dual role in tumor suppression and oncogenesis. Discovered in 1997. TP73 shares structural and functional similarities with p53. including a transactivation domain, DNA-binding domain, and oligomerization domain. It regulates cell cycle arrest, apoptosis, and DNA repair in response to stress signals. Unlike p53. TP73 is rarely mutated in cancers but exhibits complex regulation through alternative promoter usage and splicing, generating two major isoforms: TAp73 (pro-apoptotic) and ΔNp73 (anti-apoptotic). These isoforms often antagonize each other, creating a balance critical for development and homeostasis.

TP73 is essential for neurodevelopment, immunity, and sensory functions. Dysregulation of TP73 isoforms contributes to cancer progression, chemoresistance, and metastasis.
