背景信息

Transducer of Regulated CREB (cAMP response element-binding protein) activity 2 (TORC2), also known as CRTC2. is a coactivator protein critical for modulating CREB-mediated transcriptional regulation. It belongs to the TORC family (TORC1. TORC2. TORC3) and plays a pivotal role in energy homeostasis, gluconeogenesis, and stress response pathways. TORC2 is primarily expressed in the liver, pancreas, and adipose tissues. Structurally, it contains conserved N-terminal and C-terminal domains that facilitate interactions with CREB and other transcription factors.

Under low-energy conditions, TORC2 is dephosphorylated and translocates to the nucleus, where it binds CREB to activate target genes like *PGC-1α* and *G6Pase*, driving glucose production. Its activity is tightly regulated by phosphorylation through AMP-activated protein kinase (AMPK) and salt-inducible kinases (SIKs), linking cellular energy status to metabolic adaptations. Dysregulation of TORC2 is implicated in metabolic disorders, including type 2 diabetes, due to aberrant hepatic glucose output, and in certain cancers via CREB-dependent oncogenic signaling.

Recombinant human TORC2 protein is engineered for research applications, enabling studies on CREB-mediated transcription, metabolic pathways, and drug discovery. Produced using bacterial or mammalian expression systems, it retains functional domains for in vitro binding assays or structural analyses. Its use has advanced understanding of glucose metabolism regulation and potential therapeutic targeting in metabolic diseases.
