BAZ2B (Bromodomain Adjacent to Zinc finger domain protein 2B) is a chromatin-associated protein belonging to the BAZ family, characterized by conserved bromodomains and zinc finger motifs. It functions as a regulatory component of chromatin remodeling complexes, particularly the Polybromo-associated BAF (PBAF) complex, which modulates chromatin structure to influence transcriptional activity. The N-terminal bromodomain enables BAZ2B to recognize acetylated lysine residues on histones, facilitating its recruitment to acetylated chromatin regions. Its C-terminal region contains tandem zinc finger domains involved in protein-DNA or protein-protein interactions.
Research highlights BAZ2B's role in epigenetic regulation, notably in neural development, cellular differentiation, and cancer progression. It interacts with histone deacetylases (HDACs) and other chromatin modifiers to repress or activate gene expression depending on cellular context. Dysregulation of BAZ2B has been implicated in malignancies, including prostate and hepatocellular carcinoma, where it may promote tumor invasiveness. Emerging studies also suggest its involvement in neurodevelopmental disorders, potentially through aberrant chromatin remodeling affecting neuronal gene networks. Despite functional overlaps with its paralog BAZ2A, BAZ2B exhibits distinct tissue-specific expression patterns, with higher abundance in brain and testes. Current investigations aim to elucidate its mechanistic contributions to disease and explore its bromodomain as a therapeutic target.
