ERG antibodies are immunological tools used to detect the ERG protein, a member of the ETS family of transcription factors encoded by the *ERG* gene. The ERG protein plays critical roles in endothelial cell differentiation, angiogenesis, and hematopoietic development. It regulates genes involved in cell proliferation, apoptosis, and vascular remodeling. In pathology, ERG expression is closely associated with certain cancers, notably prostate cancer and vascular tumors.
In prostate cancer, ERG overexpression is frequently driven by a chromosomal translocation fusing the *TMPRSS2* promoter with the *ERG* gene (TMPRSS2-ERG), observed in approximately 50% of cases. ERG immunohistochemistry (IHC) is thus widely used as a diagnostic marker to confirm prostatic adenocarcinoma and distinguish it from other malignancies. In vascular tumors, ERG is a sensitive marker for endothelial differentiation, aiding in the diagnosis of angiosarcoma and epithelioid hemangioendothelioma.
ERG antibodies are also utilized in research to study vascular biology, tumorigenesis, and mechanisms of gene regulation. Their specificity and reliability make them valuable in both diagnostic pathology and experimental studies. However, interpretation requires caution, as ERG can be expressed in non-neoplastic endothelial cells and non-vascular tumors like Ewing sarcoma. Despite this, ERG remains a pivotal biomarker in understanding and managing cancers with ETS-family gene aberrations.
