背景信息

DDX58. also known as retinoic acid-inducible gene I (RIG-I), is a cytosolic RNA helicase critical in innate antiviral immunity. It acts as a pattern recognition receptor (PRR) that detects viral RNA, particularly short double-stranded RNA (dsRNA) or uncapped 5′-triphosphate single-stranded RNA, triggering downstream signaling to produce type I interferons (IFNs) and proinflammatory cytokines. Structurally, DDX58 contains two N-terminal caspase activation and recruitment domains (CARDs), a central DExD/H-box helicase domain, and a C-terminal regulatory domain. Upon RNA binding, DDX58 undergoes conformational changes, enabling CARD-mediated interaction with mitochondrial antiviral-signaling protein (MAVS) to activate IRF3/7 and NF-κB pathways.

Antibodies targeting DDX58 are essential tools for studying its expression, localization, and function in viral infections, autoimmune diseases, and cancer. They are widely used in techniques like Western blot, immunofluorescence, immunoprecipitation, and flow cytometry. Specific antibodies can distinguish between inactive and activated states of DDX58. aiding in research on its post-translational modifications (e.g., ubiquitination) or mutations linked to immune dysregulation. Commercially available monoclonal and polyclonal antibodies vary in species reactivity (human, mouse, etc.) and applications, requiring validation for experimental contexts. Dysregulation of DDX58 is implicated in conditions like Aicardi-Goutières syndrome and certain cancers, making its antibodies valuable for diagnostic and therapeutic exploration.
