CASC5 (Cancer Susceptibility Candidate 5), also known as KNSTRN or KNL1. is a critical protein involved in mitotic checkpoint regulation and chromosomal segregation during cell division. It functions as a core component of the kinetochore, a multi-protein structure essential for proper spindle microtubule attachment and the spindle assembly checkpoint (SAC). CASC5 interacts with other SAC proteins, such as BUB1 and MAD2. to prevent premature anaphase onset until all chromosomes achieve bipolar attachment, ensuring genomic stability.
