背景信息

The TACR1 gene encodes tachykinin receptor 1 (TACR1), also known as the neurokinin-1 receptor (NK1R), a G protein-coupled receptor (GPCR) that binds the neuropeptide substance P (SP) with high affinity. As a member of the tachykinin receptor family, TACR1 plays a central role in mediating neurogenic inflammation, pain signaling, and emotional regulation. It is widely expressed in the central and peripheral nervous systems, as well as in immune cells and epithelial tissues. Dysregulation of TACR1 signaling has been implicated in numerous pathological conditions, including chronic pain, depression, anxiety, inflammatory disorders, and cancer progression.

Recombinant TACR1 protein is engineered using heterologous expression systems (e.g., mammalian, insect, or bacterial cells) to produce purified, functional receptor proteins for structural and functional studies. This recombinant protein retains key domains required for ligand binding and intracellular signaling, enabling researchers to investigate receptor-ligand interactions, downstream signaling pathways (e.g., PLC-IP3. MAPK), and pharmacological modulation. Its production often involves epitope tagging (e.g., His-tag) for simplified purification and detection.

The development of recombinant TACR1 has advanced drug discovery efforts targeting neurokinin pathways. It serves as a critical tool for high-throughput screening of NK1R antagonists, such as aprepitant used in chemotherapy-induced nausea. Structural studies using recombinant protein have also elucidated binding mechanisms of both endogenous peptides and synthetic inhibitors, guiding the design of novel therapeutics for neurological and inflammatory diseases. Current research continues to explore its role in cancer biology and immune modulation, expanding potential clinical applications.
