Recombinant human UCHL5IP (Ubiquitin C-terminal Hydrolase L5 Interacting Protein), also known as Ubp6-associated protein or ASPSCR1. is a regulatory component of the ubiquitin-proteasome system. It interacts with UCHL5 (UCH37), a deubiquitinating enzyme (DUB) attached to the 19S regulatory cap of the 26S proteasome. UCHL5IP plays a dual role: it enhances UCHL5's hydrolase activity toward ubiquitin chains while restricting substrate specificity, ensuring precise cleavage during protein degradation. This protein is critical in maintaining proteostasis by editing ubiquitin signals and modulating proteasomal processing of polyubiquitinated substrates. Beyond its proteasomal function, UCHL5IP has been implicated in transcriptional regulation and cell cycle control.
