CCL28 (Chemokine (C-C motif) ligand 28), also known as Mucosal Epidermal Chemokine (MEC), is a small secretory protein belonging to the CC-chemokine family. It is constitutively expressed by mucosal epithelial cells in tissues such as the intestines, salivary glands, and mammary glands, playing a pivotal role in regulating immune responses at mucosal surfaces. Structurally, CCL28 contains 108 amino acids, featuring a conserved chemokine domain with four cysteine residues that stabilize its tertiary structure. Its mature form (aa 23-108) is generated by proteolytic cleavage of the signal peptide.
Functionally, CCL28 binds to two receptors: CCR10 and CCR3. This dual-receptor interaction enables it to recruit diverse immune cells, including IgA antibody-secreting plasma cells, regulatory T cells, and eosinophils, to mucosal sites. It contributes to antimicrobial defense by directly suppressing microbial growth and enhancing adaptive immunity. Additionally, CCL28 is implicated in tissue-specific homing of lymphocytes, inflammation modulation, and wound healing.
Recombinant CCL28 is produced via heterologous expression systems (e.g., E. coli or mammalian cells) followed by purification using chromatography techniques. Its applications span in vitro and in vivo studies exploring mucosal immunity, allergic diseases, and cancer metastasis. As a research tool, it aids in elucidating chemotaxis mechanisms and testing therapeutic strategies targeting CCR10/CCR3 pathways. However, recombinant versions may lack post-translational modifications present in native proteins, necessitating functional validation for specific experimental contexts.
