MNAT1 (menin-MAML complex subunit-associated protein 1), also known as CDK-activating kinase assembly factor MAT1. is a critical regulatory protein involved in cell cycle progression, transcriptional regulation, and chromatin modification. It functions as a component of the CDK-activating kinase (CAK) complex, which includes CDK7. cyclin H, and MNAT1. This complex plays a dual role: it regulates the activity of RNA polymerase II by phosphorylating its C-terminal domain (CTD) during transcription initiation, and it activates cyclin-dependent kinases (CDKs) essential for cell cycle transitions, particularly G1/S and G2/M checkpoints.

MNAT1 acts as an assembly scaffold, stabilizing interactions between CDK7 and cyclin H, thereby enhancing CAK kinase activity. Its structural domains, including a conserved RING finger and a hydrophobic region, mediate protein-protein interactions critical for these functions. Beyond transcription and cell cycle control, MNAT1 is implicated in DNA repair and apoptosis, interacting with pathways like transcription-coupled repair (TCR).
