背景信息

**Background of Recombinant Human CSEN Protein**

CSEN, also known as DRE antagonist modulator (DREAM), is a calcium-sensing protein belonging to the neuronal calcium sensor (NCS) family. Encoded by the *KCNIP3* gene in humans (located on chromosome 19p13.12), CSEN contains four EF-hand domains that bind calcium ions, enabling its role in calcium-mediated signaling. It functions as a transcriptional repressor, regulating gene expression by binding to downstream regulatory element (DRE) sites in target DNA.

In the nervous system, CSEN is highly expressed and implicated in neuronal excitability, synaptic plasticity, and apoptosis. It interacts with voltage-gated potassium channels (e.g., Kv4) to modulate action potentials and with presenilin to influence γ-secretase activity, linking it to Alzheimer’s disease pathways. CSEN also represses pro-apoptotic genes like *Bax*, highlighting its role in cell survival. Dysregulation of CSEN is associated with neurodegenerative disorders, including Alzheimer’s and Parkinson’s diseases, as well as cardiac arrhythmias.

Recombinant human CSEN protein is typically produced via bacterial or mammalian expression systems for biochemical studies. It serves as a critical tool for investigating calcium signaling, neuroprotection, and transcriptional regulation, offering insights into therapeutic strategies targeting neurodegenerative conditions and calcium-related pathologies.
