背景信息

Recombinant human CREB3L4 (cAMP responsive element binding protein 3-like 4) is a transcription factor belonging to the CREB/ATF family. It shares structural homology with other CREB3 subfamily members, characterized by a basic leucine zipper (bZIP) domain for DNA binding and dimerization. Unlike its paralogs, CREB3L4 is primarily expressed in secretory tissues such as the prostate, pancreas, and gastrointestinal tract, where it localizes to the endoplasmic reticulum (ER) or Golgi apparatus under normal conditions.

Functionally, CREB3L4 is activated through regulated intramembrane proteolysis (RIP) in response to cellular stress or specific signals. Cleavage releases its N-terminal domain, which translocates to the nucleus to regulate genes involved in protein secretion, ER stress responses, and cellular proliferation. Studies link CREB3L4 to cancer progression, particularly hormone-sensitive cancers (e.g., prostate and breast), where its overexpression correlates with tumor aggressiveness and therapy resistance.
