Bromodomain-containing protein 7 (BRD7) is a member of the bromodomain protein family, characterized by conserved structural motifs that recognize acetylated lysine residues on histones, facilitating interactions with chromatin. Initially identified as a tumor suppressor, BRD7 plays critical roles in transcriptional regulation, chromatin remodeling, and cellular processes such as cell cycle control, differentiation, and apoptosis. Structurally, it contains two N-terminal bromodomains and a C-terminal domain involved in protein interactions. BRD7 interacts with components of the SWI/SNF chromatin-remodeling complex and serves as a co-regulator for transcription factors, including p53. modulating target genes linked to tumor suppression. Dysregulation of BRD7 has been implicated in cancer progression, notably in nasopharyngeal carcinoma, breast cancer, and glioblastoma, where its loss or mutation correlates with enhanced proliferation, metastasis, and genomic instability. Beyond oncology, BRD7 participates in inflammatory responses, autophagy, and metabolic regulation.
