背景信息

Re MAL-like protein (MALL), also known as MAL2. is a member of the MAL proteolipid family involved in membrane trafficking and cell polarization. It shares structural homology with MAL (T-cell differentiation protein), featuring four transmembrane domains (TMDs) and cytoplasmic N- and C-termini. MALL is ubiquitously expressed, prominently in epithelial cells, and localizes to cholesterol-rich lipid rafts, facilitating cargo sorting and vesicular transport between apical membranes and endosomes.

Functionally, MALL contributes to maintaining epithelial polarity by regulating apical protein delivery. Emerging studies link it to clathrin-independent endocytosis, particularly in hepatocyte bile transport. Dysregulation of MALL is implicated in tumorigenesis, showing altered expression in cancers like hepatocellular carcinoma and breast cancer. Overexpression may promote metastasis via membrane protein trafficking pathways, while downregulation disrupts cell-cell adhesion. Its role in cancer remains underexplored, but MALL is proposed as a potential biomarker or therapeutic target. Recent proteomic studies also suggest interactions with signaling proteins like EGFR, hinting at broader roles in cellular signaling. Despite progress, mechanistic details of MALL's trafficking regulation and disease associations require further validation, particularly in vivo models.
