背景信息

CADM3 (Cell Adhesion Molecule 3), also known as Necl-3 or SynCAM3. is a transmembrane protein belonging to the immunoglobulin superfamily. It plays critical roles in cell-cell adhesion, synaptic organization, and intracellular signaling. Structurally, CADM3 contains three extracellular immunoglobulin-like domains, a single transmembrane region, and a cytoplasmic tail that interacts with scaffolding proteins like CASK. It is predominantly expressed in the nervous system, where it facilitates synapse formation, axonal guidance, and myelination by mediating homophilic or heterophilic interactions with other adhesion molecules like CADM1 or CADM2.

Recombinant CADM3 protein is engineered using expression systems (e.g., HEK293 cells, E. coli) to produce soluble forms of its extracellular domain for functional studies. This recombinant protein retains binding capacity, enabling researchers to investigate its role in neural development, tumor suppression, and immune modulation. Notably, CADM3 has been implicated in cancer biology, as its downregulation is associated with metastasis in certain malignancies. In neurodegenerative contexts, altered CADM3 expression correlates with synaptic dysfunction. Researchers utilize recombinant CADM3 to explore therapeutic targets, develop diagnostic tools, or model protein-protein interaction networks in vitro. Its applications span neurobiology, oncology, and regenerative medicine, offering insights into disease mechanisms and potential interventions.
