背景信息

CADM1 (Cell Adhesion Molecule 1), also known as SynCAM1 or IGSF4. is a transmembrane glycoprotein belonging to the immunoglobulin (Ig) superfamily. It plays a critical role in cell-cell adhesion, synaptic organization, and immune regulation through homophilic or heterophilic interactions. Structurally, CADM1 contains three extracellular Ig-like domains, a transmembrane region, and a cytoplasmic tail that binds intracellular scaffolding proteins like DAL-1 and protein 4.1B, facilitating signal transduction and cytoskeletal linkage. Its expression is prominent in neurons, epithelial cells, and immune cells, where it modulates synaptic plasticity, epithelial barrier integrity, and T-cell activation.

Recombinant CADM1 protein is engineered to study these biological processes in vitro or in vivo. Typically produced in mammalian or insect cell systems to ensure proper post-translational modifications (e.g., glycosylation), the recombinant form often includes the extracellular domain for functional studies. Researchers use it to investigate CADM1’s role in neural development, tumor suppression (it acts as a tumor suppressor in multiple cancers), and immune synapse formation. Dysregulation of CADM1 is linked to neuropsychiatric disorders, metastasis, and autoimmune diseases, making its recombinant form valuable for drug screening, structural analysis, and therapeutic antibody development. Its soluble variants also serve as decoys to inhibit pathological interactions in disease models.
