背景信息

SOX8. SOX9. SOX17. and SOX18 belong to the SOX (Sry-related HMG-box) family of transcription factors, which regulate diverse developmental processes by binding DNA through a conserved high-mobility group (HMG) domain. These proteins play critical roles in cell fate determination, differentiation, and tissue homeostasis. SOX8 and SOX9 (subgroup E) are involved in sex determination, neural crest development, and chondrogenesis. SOX9 is essential for male gonad development and cartilage formation, while SOX8 exhibits overlapping functions with SOX9 in certain contexts, such as maintaining progenitor cells in the nervous system. SOX17 and SOX18 (subgroup F) are implicated in endodermal lineage specification, vascular/lymphatic development, and organogenesis. SOX17 regulates endoderm-derived organ formation (e.g., gut, liver) and pluripotency in stem cells, whereas SOX18 is crucial for lymphatic vessel formation and cardiovascular development.

Antibodies targeting these SOX proteins are vital tools for studying their expression, localization, and functional roles in development and disease. They are widely used in techniques like immunohistochemistry, Western blotting, and immunofluorescence to map protein distribution in tissues or assess expression levels in pathological conditions, such as cancer. For instance, SOX9 overexpression is linked to tumor progression in multiple cancers, while SOX17/18 dysregulation is associated with vascular anomalies or metastasis. Validation of antibody specificity is critical due to high homology among SOX family members, requiring careful controls (e.g., knockout validation) to avoid cross-reactivity. These antibodies continue to advance research in developmental biology, regenerative medicine, and oncology.
