MAT1 (Menage à Trois 1), also known as MNAT1. is a critical regulatory protein that functions as a component of the CDK-activating kinase (CAK) complex. This complex, composed of MAT1. cyclin H, and CDK7. plays dual roles in cell cycle regulation and transcriptional processes. As part of CAK, MAT1 stabilizes the interaction between CDK7 and cyclin H, enabling CDK7 to phosphorylate key cell cycle kinases (CDKs 1. 2. 4. 6) to drive cell cycle progression. Additionally, the CAK complex associates with the transcription factor IIH (TFIIH) to facilitate RNA polymerase II-mediated transcription initiation.
MAT1 antibodies are widely used in research to study its expression, localization, and interactions in cellular pathways. Dysregulation of MAT1 has been implicated in cancers, including breast and ovarian cancers, where its overexpression correlates with tumor proliferation and poor prognosis. These antibodies aid in detecting MAT1 levels in clinical samples, elucidating its role in oncogenesis and therapeutic resistance.
Studies also explore MAT1's potential as a therapeutic target, particularly in combination with CDK7 inhibitors. Its involvement in both cell cycle control and transcription makes it a focal point for understanding diseases linked to proliferative or transcriptional abnormalities. MAT1 antibodies thus serve as essential tools for dissecting molecular mechanisms and developing targeted cancer therapies.
