背景信息

CD200R2 (CD200 Receptor 2) is a cell surface glycoprotein belonging to the immunoglobulin (Ig) superfamily, primarily involved in immune regulation. It shares structural homology with CD200R1. another receptor for CD200 (OX2), a membrane-bound protein expressed on various cell types. Unlike CD200R1. which contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) and mediates immunosuppressive signals, CD200R2 lacks canonical ITIMs, suggesting a distinct functional role. CD200R2 is expressed on myeloid cells, including macrophages and dendritic cells, and may modulate inflammatory responses through alternative signaling pathways.

The CD200-CD200R axis is critical for maintaining immune homeostasis by dampening excessive immune activation. CD200R2’s exact mechanism remains less defined, but studies propose it may act as a decoy receptor, compete with CD200R1 for ligand binding, or initiate unique downstream signals to fine-tune immune tolerance. Its involvement in autoimmune diseases, cancer, and infections highlights therapeutic potential. For instance, dysregulated CD200R2 expression correlates with chronic inflammation and tumor immune evasion.

Recombinant CD200R2 proteins are engineered to study receptor-ligand interactions, signaling, and therapeutic targeting. Typically produced in mammalian systems (e.g., HEK293 cells) to ensure proper glycosylation, these proteins retain the extracellular domain for functional studies. Applications include blocking assays, immune cell modulation experiments, and drug development. Research on CD200R2 may unveil novel strategies to treat inflammatory disorders or enhance immunotherapy efficacy by manipulating immune checkpoint pathways. However, further studies are needed to fully elucidate its biological significance and translational value.
