Interleukin-11 receptor alpha 1 (IL11RA1) is a key component of the interleukin-11 (IL-11) receptor complex, which mediates signaling of the cytokine IL-11. This transmembrane protein belongs to the type I cytokine receptor family and shares structural homology with other IL-6 receptor family members. IL11RA1 pairs with glycoprotein 130 (gp130) to form a functional receptor capable of activating downstream signaling pathways, including JAK/STAT, MAPK, and PI3K, upon IL-11 binding.
Recombinant IL11RA1 protein is engineered to mimic the extracellular domain of the native receptor, often produced in mammalian or bacterial expression systems with purification tags (e.g., His-tag) for research applications. Its production enables studies of IL-11/IL11RA1 interactions, receptor activation mechanisms, and competitive inhibition experiments. Dysregulation of IL-11 signaling is implicated in pathological conditions such as fibrosis, inflammatory diseases, osteoporosis, and cancer progression, making recombinant IL11RA1 a valuable tool for investigating therapeutic interventions.
Notably, IL11RA1's role in tissue homeostasis and repair has spurred interest in its soluble form (sIL11RA), which may act as a decoy receptor to modulate IL-11 activity. Studies using recombinant IL11RA1 have clarified its tissue-specific functions, including its contribution to hematopoietic stem cell regulation, bone metabolism, and epithelial cell survival. Emerging evidence also links IL11RA1 mutations to craniofacial developmental disorders like Crouzon syndrome, highlighting its importance in skeletal development. As IL-11 targeting therapies gain traction, recombinant IL11RA1 remains critical for drug screening, structural studies, and elucidating disease mechanisms.
