背景信息

ZBP1 (Z-DNA-binding protein 1), also known as DAI (DNA-dependent activator of interferon regulatory factors) or DLM-1. is a cytosolic nucleic acid sensor critical in innate immune responses. It belongs to the Zα domain-containing protein family and is characterized by its ability to recognize non-canonical left-handed Z-form nucleic acids (Z-DNA/Z-RNA), which are often associated with viral infections or cellular stress. Structurally, ZBP1 contains two N-terminal Zα domains that mediate Z-nucleic acid binding and two RIP homotypic interaction motifs (RHIM) at the C-terminus, enabling interactions with other RHIM-containing proteins like RIPK3 and RIPK1.

Initially identified as a tumor-associated protein, ZBP1 gained prominence for its role in antiviral defense. It detects viral nucleic acids, particularly during infections by DNA viruses (e.g., herpesviruses) or RNA viruses (e.g., influenza), triggering downstream signaling pathways that activate NF-κB and interferon regulatory factors (IRFs). This leads to the production of type I interferons (IFNs) and pro-inflammatory cytokines, amplifying antiviral responses. Beyond immunity, ZBP1 is a key mediator of programmed cell death pathways. Upon sensing Z-RNA during viral infection or cellular damage, it activates RIPK3-dependent necroptosis or apoptosis through caspase-8. depending on cellular context. Dysregulation of ZBP1 signaling has been implicated in inflammatory diseases, autoimmune disorders, and cancer progression.

Recombinant ZBP1 protein, typically produced in bacterial or mammalian expression systems, is widely used to study its structural features, ligand interactions, and signaling mechanisms. It serves as a tool for elucidating its dual roles in host defense and pathology, as well as for developing therapeutic strategies targeting ZBP1-mediated pathways in infectious and inflammatory diseases.
