CSH1 (chorionic somatomammotropin hormone 1), also known as human placental lactogen (hPL), is a peptide hormone produced predominantly by the syncytiotrophoblast cells of the placenta during pregnancy. It belongs to the growth hormone (GH) prolactin gene family and shares structural homology with both GH (85%) and prolactin. CSH1 is encoded by the CSH1 gene located on chromosome 17q23.3 within a cluster of five closely related genes, including GH and placental lactogen variants.
Functionally, CSH1 plays critical roles in maternal metabolic adaptation and fetal development. It induces insulin resistance in pregnant individuals, redirecting glucose toward the fetus while promoting lipolysis to provide alternative energy sources for the mother. Additionally, it stimulates mammary gland development and works synergistically with progesterone to maintain pregnancy. CSH1 levels rise progressively during gestation, correlating with placental mass, making it a clinical marker for placental function assessment.
Recombinant CSH1 protein is produced using biotechnological platforms such as bacterial (E. coli), yeast, or mammalian cell expression systems. The production typically involves gene cloning, protein expression, and purification steps like affinity chromatography. Recombinant versions retain the native hormone's biological activities while offering standardized quality and reduced batch variability compared to placenta-derived extracts.
Research applications include studying pregnancy-related metabolic disorders (e.g., gestational diabetes), fetal growth regulation mechanisms, and developing therapeutic strategies for placental insufficiency. Its mitogenic and metabolic effects are also explored in oncology and endocrinology research. As a recombinant protein, CSH1 provides a crucial tool for both basic research and potential clinical interventions in obstetrical complications.
