背景信息

CCL16 (C-C motif chemokine ligand 16), also known as HCC-4 or LEC, is a member of the CC chemokine family encoded by the SCYA16 gene in humans. It is constitutively expressed in the liver, spleen, and thymus, with induced expression observed during inflammatory responses. This 12-14 kDa protein contains a conserved N-terminal motif critical for receptor interaction and undergoes proteolytic processing to generate bioactive isoforms. As a recombinant protein, CCL16 is typically produced in Escherichia coli or mammalian expression systems, ensuring high purity and endotoxin-free properties for research applications.

Functionally, CCL16 acts as a chemoattractant through binding to G protein-coupled receptors CCR1. CCR2. CCR5. and CCR8. mediating immune cell migration and activation. It recruits monocytes, lymphocytes, dendritic cells, and neutrophils to sites of inflammation or injury. Beyond its role in innate immunity, CCL16 participates in angiogenesis, wound healing, and tissue remodeling by interacting with endothelial cells. Its extended C-terminal domain enables unique interactions with glycosaminoglycans, influencing chemotactic gradient formation.

Dysregulation of CCL16 has been implicated in pathological conditions including rheumatoid arthritis, atherosclerosis, and HIV infection. In cancer biology, it exhibits dual roles – promoting tumor progression through angiogenesis in hepatocellular carcinoma while potentially suppressing metastasis in breast cancer via immune modulation. Recombinant CCL16 serves as a vital tool for studying leukocyte trafficking mechanisms, screening anti-inflammatory compounds, and developing diagnostic biomarkers for chronic inflammatory diseases. Current research explores its therapeutic potential in modulating immune responses and tissue repair processes.
