背景信息

CD86. also known as B7-2. is a transmembrane glycoprotein belonging to the B7 family of co-stimulatory molecules. It is primarily expressed on antigen-presenting cells (APCs) such as dendritic cells, macrophages, and activated B cells. CD86 interacts with CD28 and CTLA-4 receptors on T cells, playing a critical role in modulating T-cell activation and immune responses. Binding to CD28 delivers a co-stimulatory signal essential for full T-cell activation, while engagement with CTLA-4 (cytotoxic T-lymphocyte-associated protein 4) suppresses T-cell activity, maintaining immune tolerance and preventing excessive inflammation. This dual regulatory function positions CD86 as a key immune checkpoint molecule involved in autoimmune diseases, transplant rejection, and cancer immunotherapy.

Recombinant CD86 proteins are engineered to study these interactions and develop therapeutic strategies. Typically produced in mammalian expression systems (e.g., CHO or HEK293 cells), recombinant CD86 retains functional extracellular domains and undergoes proper post-translational modifications, ensuring structural and functional fidelity. Some constructs fuse the Fc region of immunoglobulins to enhance stability and facilitate detection in assays. These proteins are widely used in vitro to investigate T-cell/APC communication, screen immune-modulating drugs (e.g., CTLA-4 inhibitors like ipilimumab), and model diseases such as rheumatoid arthritis or graft-versus-host disease. Additionally, they serve as critical tools in developing bispecific antibodies or CAR-T therapies targeting immune checkpoints. By enabling precise control over co-stimulatory signals, recombinant CD86 accelerates research into balancing immune activation and tolerance for therapeutic applications.
