背景信息

Kallikrein-related peptidase 10 (KLK10), also known as the normal epithelial cell-specific 1 (NES1) protein, is a member of the tissue kallikrein serine protease family, which comprises 15 secreted enzymes (KLK1-KLK15) with diverse roles in physiological and pathological processes. KLK10 is encoded by the KLK10 gene located on chromosome 19q13.4 and is primarily expressed in hormone-regulated tissues, including the breast, prostate, ovary, and skin. It functions as a trypsin-like protease, involved in extracellular matrix remodeling, cellular signaling, and inflammation regulation through cleavage of specific substrates.

Originally identified as a tumor suppressor, KLK10 is frequently downregulated in cancers such as ovarian, breast, and prostate due to promoter hypermethylation. Reduced KLK10 expression correlates with poor prognosis, suggesting its role in inhibiting metastasis and angiogenesis. Paradoxically, some studies report KLK10 overexpression in certain malignancies, indicating context-dependent roles influenced by tissue microenvironment, post-translational modifications, or interactions with other kallikreins.

Recombinant KLK10 protein is produced using expression systems like mammalian cells or bacteria, enabling studies on its biochemical properties and therapeutic potential. Purified KLK10 is utilized to investigate protease-activated receptor signaling, biomarker validation, and its dualistic behavior in cancer progression. Additionally, it serves as a reagent for developing diagnostic assays or inhibitors targeting dysregulated kallikrein activity in diseases. Ongoing research aims to clarify its mechanistic duality and harness its diagnostic or therapeutic value in precision oncology.
