**Background of DEK Antibodies**
DEK antibodies are autoantibodies targeting the DEK protein, a nuclear factor encoded by the *DEK* gene. DEK is involved in chromatin remodeling, transcriptional regulation, and DNA repair, playing roles in cell proliferation and differentiation. Initially identified in acute myeloid leukemia (AML) as part of a fusion protein (DEK-CAN), DEK gained attention in autoimmunity when DEK autoantibodies were detected in patients with systemic lupus erythematosus (SLE) and other rheumatic diseases.
These antibodies are predominantly associated with juvenile idiopathic arthritis (JIA) and SLE, particularly in subsets with articular or cutaneous manifestations. DEK autoantibodies are detected via ELISA or immunoblot, often alongside other antinuclear antibodies (ANAs). Their presence correlates with specific clinical features, such as synovitis in JIA, though their pathogenic role remains unclear.
DEK is highly immunogenic due to post-translational modifications (e.g., citrullination) in inflammatory environments, potentially exposing cryptic epitopes. Research suggests DEK antibodies may contribute to disease by forming immune complexes or interfering with DEK’s regulatory functions. However, their diagnostic utility is less established compared to classical markers (e.g., anti-dsDNA). Current studies focus on elucidating DEK’s role in autoimmune pathways and its potential as a therapeutic target or biomarker for disease stratification.
