Beta-2 microglobulin (B2M) is a 12 kDa protein component of the major histocompatibility complex (MHC) class I molecule, expressed on nearly all nucleated cells. It plays a critical role in immune surveillance by stabilizing MHC-I structure and presenting antigenic peptides to CD8+ T cells. B2M is constitutively shed into biological fluids, with serum and urine levels serving as biomarkers for renal dysfunction, inflammatory diseases, and certain cancers. Elevated B2M correlates with conditions like multiple myeloma, chronic kidney disease, and autoimmune disorders.
Antibodies targeting B2M are essential tools in research and diagnostics. Monoclonal and polyclonal anti-B2M antibodies are widely used in immunoassays (e.g., ELISA, Western blot), immunohistochemistry, and flow cytometry to quantify B2M expression, assess MHC-I surface localization, or study disease mechanisms. In clinical settings, they aid in monitoring B2M levels for disease progression or treatment response. Therapeutic applications are also explored, including blocking B2M to modulate immune responses in transplantation or cancer immunotherapy. However, challenges remain, such as ensuring antibody specificity given B2M’s structural similarity to other immunoglobulin domains. Recent advances include recombinant antibodies engineered for enhanced affinity, supporting precision in both basic research and translational studies.
