背景信息

Glycophorin C (GPC), encoded by the *GYPC* gene, is a minor sialoglycoprotein expressed on the surface of human erythrocytes. It plays a critical role in maintaining membrane mechanical stability by interacting with the cytoskeletal protein 4.1R. GPC is also a carrier for the Gerbich blood group antigens (Ge), which include high-frequency (Ge2. Ge3. Ge4) and low-frequency (Ge1. Ge5–Ge13) epitopes. Antibodies targeting Glycophorin C are primarily relevant in transfusion medicine and hematology. Naturally occurring anti-Ge antibodies are rare but can cause hemolytic transfusion reactions or hemolytic disease of the fetus/newborn (HDFN) in individuals lacking specific Gerbich antigens. Clinically significant anti-GPC antibodies are typically IgG and may arise following exposure through transfusion or pregnancy. In research, GPC antibodies are utilized to study red cell membrane disorders, such as hereditary elliptocytosis or Southeast Asian ovalocytosis, where GPC-4.1R interactions are disrupted. Monoclonal anti-GPC antibodies (e.g., BRIC4. BRIC10) are commonly employed in blood typing, immunoassays, and flow cytometry to characterize erythrocyte membrane integrity or diagnose Ge antigen variants. Recent studies also explore GPC's role as a receptor for *Plasmodium falciparum* erythrocyte-binding antigen 140 (EBA-140) in malaria pathogenesis, highlighting its broader biomedical significance.
