背景信息

The Rad9 protein is a conserved component of the DNA damage response (DDR) machinery, primarily known for its role in maintaining genome stability. In eukaryotic cells, Rad9 forms the 9-1-1 complex with Rad1 and Hus1. a heterotrimeric clamp structurally analogous to PCNA. This complex is loaded onto DNA lesions, such as double-strand breaks or stalled replication forks, to activate checkpoint signaling pathways. Rad9 facilitates the recruitment of effector kinases like Chk1 and Chk2. which halt the cell cycle to allow repair or trigger apoptosis if damage is irreparable. It also interacts with DNA repair proteins, including those involved in homologous recombination and nucleotide excision repair.

Rad9 antibodies are critical tools for studying DDR mechanisms. They are used in techniques like Western blotting, immunofluorescence, and co-immunoprecipitation to detect Rad9 expression, localization, and protein interactions. Phospho-specific antibodies can identify post-translational modifications (e.g., phosphorylation) that regulate Rad9 activity during stress responses. Research applications span cancer biology, as dysregulated DDR pathways contribute to tumorigenesis and therapy resistance, and neurodegeneration, where defective DNA repair links to disease progression. Additionally, studies in model organisms (e.g., yeast, mice) using Rad9 antibodies have elucidated evolutionary conservation of DDR pathways. These antibodies are essential for dissecting how Rad9 coordinates cell cycle arrest, DNA repair, and apoptosis, offering insights into therapeutic strategies targeting genomic instability.
