背景信息

Phospho-CD3 zeta (Tyr142) antibodies specifically detect the phosphorylated form of the CD3ζ (CD3 zeta) chain at tyrosine residue 142. a critical post-translational modification in T-cell receptor (TCR) signaling. The CD3ζ chain is a component of the CD3 complex, which associates with the TCR to form the TCR-CD3 complex essential for antigen recognition and T-cell activation. Upon TCR engagement with antigen-MHC complexes, immunoreceptor tyrosine-based activation motifs (ITAMs) within CD3ζ become phosphorylated by Src family kinases (e.g., Lck). Phosphorylation at Tyr142. located in the third ITAM domain, facilitates recruitment and activation of downstream signaling molecules like ZAP-70. initiating signaling cascades that drive T-cell activation, proliferation, and cytokine production. Dysregulation of CD3ζ phosphorylation is implicated in immune disorders, T-cell exhaustion, and resistance to cancer immunotherapy. Antibodies targeting phospho-CD3ζ (Tyr142) are widely used in research to study TCR signaling dynamics, T-cell activation status, and therapeutic responses (e.g., checkpoint inhibitors). They enable detection of this transient modification via techniques like flow cytometry, Western blotting, or immunofluorescence, providing insights into immune cell function in autoimmunity, infections, and cancer. Specificity validation is crucial, as cross-reactivity with other phosphorylated ITAM residues may occur.
