NAIP (Neuronal Apoptosis Inhibitory Protein) is a member of the inhibitor of apoptosis (IAP) protein family, initially identified for its role in preventing programmed cell death in neurons. In humans, the NAIP gene is part of a cluster on chromosome 5q13.2. encoding a protein characterized by baculovirus IAP repeat (BIR) domains and a nucleotide-binding domain (NOD). Functionally, NAIP has evolved beyond apoptosis regulation to play a critical role in innate immunity. It acts as a cytosolic sensor for bacterial components, particularly flagellin and type III secretion system (T3SS) proteins of intracellular pathogens like *Salmonella* and *Legionella*.
Upon ligand recognition, NAIP interacts with NLRC4 (NOD-like receptor family CARD domain-containing protein 4) to form the NAIP-NLRC4 inflammasome. This complex activates caspase-1. triggering pyroptosis (inflammatory cell death) and the release of pro-inflammatory cytokines IL-1β and IL-18 to combat infection. Genetic variations in NAIP are linked to susceptibility to infections and inflammatory disorders. NAIP-specific antibodies are vital tools for studying its expression, inflammasome assembly, and pathogen-host interactions. They enable applications in immunoblotting, immunofluorescence, and flow cytometry, aiding research on immune signaling pathways and therapeutic targeting of inflammasome-related diseases. Dysregulation of NAIP has been implicated in autoinflammatory conditions and cancer, highlighting its dual role in cell survival and immune defense.
