HOMER1 antibodies target proteins encoded by the *HOMER1* gene, a member of the HOMER family of scaffolding proteins involved in synaptic organization and signaling. HOMER1 isoforms (HOMER1a, HOMER1b/c) regulate glutamatergic neurotransmission by binding to metabotropic glutamate receptors (mGluRs), inositol trisphosphate receptors (IP3Rs), and other postsynaptic density (PSD) proteins. The long isoforms (HOMER1b/c) constitutively form multimers, stabilizing synaptic complexes, while the short isoform (HOMER1a), an immediate-early gene product induced by neuronal activity, disrupts these complexes to modulate synaptic plasticity.
HOMER1 is critical for neurodevelopmental processes, learning, and memory. Dysregulation has been linked to neurological and psychiatric disorders, including schizophrenia, Alzheimer’s disease, and epilepsy. In cancer, HOMER1 overexpression is associated with tumor progression and metastasis.
HOMER1 antibodies are widely used in neuroscience and oncology research to study protein expression, localization, and interactions via techniques like Western blot, immunohistochemistry, and immunoprecipitation. Commercial antibodies vary in specificity, often distinguishing between isoforms (e.g., HOMER1a vs. HOMER1b/c). Validation is essential, as cross-reactivity with HOMER2/HOMER3 may occur. These tools help elucidate HOMER1's role in synaptic pathology, calcium signaling, and disease mechanisms.
