Ccl12 (C-C motif chemokine ligand 12), also known as monocyte chemoattractant protein-5 (MCP-5), is a small secreted protein belonging to the CC chemokine family. It functions as a chemoattractant for immune cells, including monocytes, T lymphocytes, eosinophils, and dendritic cells, by binding to specific receptors such as CCR2 and CCR3. Ccl12 plays a critical role in mediating inflammatory responses, immune surveillance, and leukocyte recruitment during infection, tissue injury, or autoimmune disorders. Its expression is upregulated in various pathological conditions, including asthma, fibrosis, atherosclerosis, and cancer, where it contributes to disease progression by promoting chronic inflammation, angiogenesis, or immune cell infiltration into tissues.
Recombinant Ccl12 protein is produced using engineered expression systems (e.g., E. coli or mammalian cells) to ensure high purity and bioactivity. The mature protein typically consists of ~75 amino acids, with a molecular weight of approximately 8-10 kDa, and features conserved CC chemokine structural motifs, including disulfide bonds critical for receptor interaction. Researchers utilize recombinant Ccl12 in vitro and in vivo to study chemotaxis, immune cell migration, and signaling pathways. It is also employed to model inflammatory diseases, evaluate potential therapeutic inhibitors, or explore its dual role in both promoting pathology (e.g., tumor metastasis) and facilitating tissue repair. Despite structural and functional similarities to Ccl2 (MCP-1), Ccl12 exhibits distinct receptor affinities and context-dependent roles, underscoring its unique relevance in immunology and disease mechanisms. Its recombinant form remains a vital tool for dissecting chemokine-mediated processes and developing targeted therapies.
