背景信息

Transglutaminase 2 (TGM2) antibodies target the TGM2 enzyme, a multifunctional protein belonging to the transglutaminase family. TGM2 catalyzes calcium-dependent post-translational protein modifications, including crosslinking, deamidation, and GTP-binding activities, playing roles in cell adhesion, apoptosis, wound healing, and extracellular matrix stabilization. It is ubiquitously expressed in tissues, with elevated levels observed in inflammatory, fibrotic, and neoplastic conditions.

Anti-TGM2 antibodies are clinically significant in autoimmune disorders. In celiac disease, autoantibodies against TGM2 (specifically tissue transglutaminase, tTG) are diagnostic markers, formed due to gluten-triggered immune responses where TGM2 deamidates gliadin peptides, enhancing their immunogenicity. These antibodies contribute to intestinal damage via immune complex deposition.

Beyond autoimmunity, TGM2 antibodies are research tools. Monoclonal antibodies help study TGM2’s roles in cancer progression, metastasis, and drug resistance, as TGM2 overexpression in tumors promotes survival signaling and extracellular matrix remodeling. Therapeutic antibodies targeting TGM2 are explored for inhibiting pathological crosslinking in fibrosis or blocking oncogenic signaling.

However, challenges remain, including TGM2’s pleiotropic functions and isoform diversity, which complicate antibody specificity and therapeutic outcomes. Research continues to optimize antibody design for precision in clinical and experimental applications.
