MLST8 (mammalian lethal with SEC13 protein 8), also known as GβL, is a critical regulatory component of the mammalian target of rapamycin (mTOR) signaling pathway. It serves as an essential scaffolding protein in three distinct complexes: mTOR complex 1 (mTORC1), mTORC2. and the GATOR2 complex (GSE). MLST8 stabilizes these complexes by mediating protein-protein interactions, particularly through its WD40 repeat domains. In mTORC1 and mTORC2. MLST8 directly binds to mTOR kinase, facilitating structural integrity and substrate recognition. While mTORC1 regulates cell growth in response to nutrients and energy status, mTORC2 controls cell survival and cytoskeletal organization. MLST8’s interaction with the GATOR2 complex further links it to amino acid sensing and mTORC1 regulation. Recombinant human MLST8 protein, produced via expression systems like E. coli or mammalian cells, retains these functional properties and is widely used in biochemical studies to dissect mTOR signaling mechanisms.
