Leiomodin 1 (LMOD1) is an actin-binding protein predominantly expressed in smooth muscle and skeletal muscle cells. As a member of the leiomodin/whirlin protein family, it plays a critical role in regulating actin filament assembly and stabilization. LMOD1 functions as an actin nucleator, promoting the formation of elongated actin filaments by binding to tropomyosin and enhancing the polymerization of actin monomers. Its structural organization includes N-terminal leucine-rich repeats and a C-terminal actin-binding domain, enabling interactions with cytoskeletal components.
LMOD1 is essential for maintaining sarcomere integrity in muscle tissues. Studies link LMOD1 mutations to nemaline myopathy and congenital muscular dystrophy, highlighting its importance in muscle development and function. Recombinant human LMOD1 protein, produced via bacterial or eukaryotic expression systems, enables in vitro exploration of its biochemical properties, interaction partners, and disease mechanisms. This engineered protein serves as a tool for studying actin dynamics, myofibril organization, and potential therapeutic interventions. Challenges in recombinant production include preserving post-translational modifications and ensuring proper folding for functional studies. Current research focuses on clarifying its regulatory pathways and leveraging recombinant LMOD1 to model muscle disorders or screen drugs targeting actin-related pathologies.
