背景信息

Recombinant human LCE5A (Late Cornified Envelope 5A) protein is a genetically engineered variant of the naturally occurring LCE5A protein, which belongs to the large family of late cornified envelope (LCE) proteins. These proteins are critical components of the epidermal differentiation complex (EDC) on chromosome 1q21. playing essential roles in skin barrier formation, keratinocyte differentiation, and response to environmental stressors. LCE5A is specifically expressed in the epidermis and is involved in the final stages of cornification, a process where keratinocytes form a protective cornified envelope to maintain skin integrity and hydration.

Structurally, LCE5A shares conserved features with other LCE proteins, including a central domain rich in proline and glycine residues, which may contribute to its cross-linking properties during epidermal maturation. Its expression is regulated by inflammatory cytokines and pathogenic stimuli, linking it to skin disorders such as psoriasis, atopic dermatitis, and ichthyosis. Genetic studies have identified associations between LCE gene cluster deletions (including LCE5A) and susceptibility to psoriasis, highlighting its pathophysiological relevance.

The recombinant form of LCE5A is typically produced in bacterial or mammalian expression systems, enabling researchers to study its biochemical properties, interactions with skin microbiota, and role in inflammatory pathways. Its applications span in vitro disease modeling, drug screening for dermatological therapies, and elucidating mechanisms of skin barrier dysfunction. Ongoing research aims to explore its potential as a therapeutic target or biomarker in chronic inflammatory skin diseases.
