背景信息

The mouse p27Kip1 (C-term T197) antibody is designed to specifically detect the phosphorylated form of p27Kip1 at threonine 197 (Thr197) near its C-terminal region. p27Kip1. encoded by the CDKN1B gene, is a cyclin-dependent kinase (CDK) inhibitor that regulates cell cycle progression by binding to and inhibiting cyclin E/CDK2 and cyclin D/CDK4 complexes, thereby blocking G1-to-S phase transition. Phosphorylation at Thr197. mediated by kinases such as AKT or RSK, promotes p27Kip1 cytoplasmic relocalization and protein stability by preventing its ubiquitin-mediated proteasomal degradation. This post-translational modification is associated with cell cycle arrest, cell motility, and tumor progression in certain cancers. The antibody is commonly used to study p27Kip1 regulation in cell cycle checkpoints, differentiation, and cancer biology. Its specificity for the phosphorylated Thr197 epitope allows researchers to distinguish the active, modified form of p27Kip1 from its unphosphorylated state in applications like Western blotting, immunoprecipitation, and immunofluorescence. Validation typically includes testing in p27Kip1-deficient cells or competition assays with phosphorylated peptides. This antibody is particularly valuable in exploring mechanisms underlying cell cycle dysregulation in mouse models of disease.
