The PBRM1 (Polybromo-1) antibody is a tool used to detect the PBRM1 protein, a key subunit of the PBAF (SWI/SNF-B) chromatin remodeling complex. PBRM1. also known as BAF180. plays a critical role in regulating gene expression by modifying chromatin structure to enable transcriptional activation or repression. It is implicated in diverse cellular processes, including DNA repair, cell cycle control, and differentiation. PBRM1 is widely recognized as a tumor suppressor, with its loss-of-function mutations frequently observed in cancers, most notably in clear cell renal cell carcinoma (ccRCC), where it is the second most commonly mutated gene after VHL. Dysregulation of PBRM1 has also been linked to other malignancies, such as bladder, lung, and liver cancers.
Antibodies targeting PBRM1 are essential for research and diagnostic applications, including immunohistochemistry (IHC), western blotting, and immunofluorescence, to assess protein expression and localization. In cancer studies, these antibodies help investigate PBRM1's role in tumorigenesis, metastasis, and therapeutic response. For example, PBRM1-deficient tumors may exhibit altered sensitivity to immunotherapy or targeted therapies. Additionally, PBRM1 antibody-based assays contribute to understanding epigenetic mechanisms and identifying potential biomarkers for prognosis or treatment stratification. However, variability in antibody specificity and validation across experimental models requires careful interpretation of results. Ongoing research aims to clarify PBRM1's context-dependent functions and therapeutic implications in precision oncology.
