The RAB27B antibody is a key tool for studying the RAB27B protein, a member of the Ras-related GTPase RAB family involved in vesicle trafficking and exocytosis. RAB27B regulates the transport and secretion of lysosome-related organelles, such as melanosomes in melanocytes, platelet dense granules, and secretory lysosomes in immune cells. It interacts with effector proteins like Slac2-a/Melanophilin and MyRIP to mediate vesicle docking and membrane fusion processes. Dysregulation of RAB27B is linked to diseases, including Griscelli syndrome type 3 (hypopigmentation disorders) and cancer metastasis, where its overexpression enhances invasive secretion in breast, prostate, and colorectal cancers.
RAB27B-specific antibodies are widely used in Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF) to detect protein expression levels, subcellular localization, and interactions in cellular models or clinical samples. These antibodies help elucidate RAB27B's role in tumor microenvironment modulation, immune evasion, and chemoresistance. Validated antibodies distinguish RAB27B from its homolog RAB27A (sharing 71% amino acid identity) through isoform-specific epitopes. Knockdown/knockout controls are often employed to confirm antibody specificity. As RAB27B emerges as a potential therapeutic target, its antibodies also support drug discovery by enabling target engagement assays and biomarker analysis in preclinical studies.
