The Mouse Leo1 antibody is a tool used to study the Leo1 protein, a conserved subunit of the RNA polymerase II-associated factor 1 (Paf1) complex in mammals. Leo1 (also known as PAFL) was initially identified in yeast and plays a critical role in transcriptional elongation, histone modification, and chromatin remodeling. In mice and humans, the Paf1 complex, comprising Leo1. Ctr9. Paf1. Cdc73. and Rtf1. regulates gene expression by interacting with RNA polymerase II and facilitating recruitment of histone-modifying enzymes, such as Set1/COMPASS for H3K4 methylation. Leo1 specifically contributes to transcriptional termination, RNA 3'-end processing, and maintenance of epigenetic marks.
Mouse Leo1 antibodies are commonly raised against recombinant or synthetic peptide antigens corresponding to conserved regions of the murine Leo1 protein. These antibodies enable detection and localization of Leo1 in techniques like Western blotting, immunoprecipitation, and chromatin immunoprecipitation (ChIP). They are particularly valuable in studying embryonic development, stem cell differentiation, and cancer biology, as dysregulation of the Paf1 complex is linked to transcriptional abnormalities in these contexts. Validated for specificity in mouse models, these antibodies help dissect Leo1's role in coordinating transcriptional and epigenetic mechanisms.
