KIR2DL4 is a member of the killer cell immunoglobulin-like receptor (KIR) family, primarily expressed on natural killer (NK) cells and a subset of T cells. Unlike most KIRs, which are either inhibitory or activating, KIR2DL4 exhibits dual functional characteristics, though it is structurally classified as an inhibitory receptor due to its immunoreceptor tyrosine-based inhibitory motif (ITIM). It uniquely interacts with human leukocyte antigen G (HLA-G), a non-classical MHC class I molecule prominently expressed in placental trophoblasts, playing a critical role in maternal-fetal immune tolerance during pregnancy.
